目的观察条件恐惧应激条件下大鼠的行为及血液相关指标变化。方法 64只Wistar大鼠随机分为恐惧应激组及空白对照组,恐惧应激组按照改良Takeuchi法制作恐惧模型大鼠,空白对照组正常饲养。检测每周(共4周)体重、自发活动相关指标、血清皮质酮和超敏C反应蛋白(hs-CRP)浓度。结果恐惧应激组大鼠的冻结行为学得分高于空白对照组(t=5.700,P<0.01);恐惧应激组体重增长减慢(第一周t=0.457,P=0.642;第二周t=2.675,P<0.05;第三周t=2.316,P<0.05;第四周t=3.930,P<0.01);自发活动实验中恐惧应激组的活动次数减少(第一周t=2.267,P<0.05;第二周t=2.990,P<0.01;第三周t=3.325,P<0.01;第四周t=2.423,P<0.05),恐惧应激组的休息时间增多(第一周t=2.621,P<0.05;第二周t=3.356,P<0.01;第三周t=2.668,P<0.05;第四周t=2.384,P<0.05);恐惧应激组血清皮质酮浓度升高(第一周t=2.231,P<0.05;第二周t=3.531,P<0.01;第三周t=2.681,P<0.05;第四周t=2.225,P<0.05);恐惧应激组血清hs-CRP浓度升高(第一周t=2.773,P<0.05;第二周t=4.043,P<0.01;第三周t=3.373,P<0.01;第四周t=3.162,P<0.01)。结论应用Takeuchi法可建立条件恐惧模型,条件恐惧应激可使机体体重增长减慢,行为和神经内分泌系统发生变化。
Objective To observe the changes of behavior and blood-related indexes of rats under conditions of fear-induced stress. Methods Sixty-four Wistar rats were randomly divided into three groups: fear stress group and blank control group. According to the modified Takeuchi method, the fear-stress group was given fear model and the blank control group was fed normally. Body weight, spontaneous activity-related indicators, serum corticosterone and hs-CRP concentrations were measured weekly (total 4 weeks). Results The scores of freezing behavior of fear-stressed rats were higher than those of the blank control group (t = 5.700, P <0.01); the weight growth of the fear-stressed rats slowed down (t = 0.457 in the first week, P = t = 2.675, P <0.05; t = 2.316 in the third week, P <0.05; t = 3.930 in the fourth week, P <0.01); the activity frequency of the fear stress group decreased in the spontaneous activity test , P <0.05; t = 2.990 in the second week, P <0.01; t = 3.325 in the third week, P <0.01; t = 2.423 in the fourth week, P < Week t = 2.621, P <0.05; t = 3.356 in the second week, P <0.01; t = 2.668 in the third week, P <0.05; t = 2.384 in the fourth week, P < (T = 2.231 in the first week, P <0.05; t = 3.531 in the second week, P <0.01; t = 2.681 in the third week, P <0.05; t = 2.225 in the fourth week, P < The concentration of hs-CRP in the stress group increased (t = 2.773 in the first week, P <0.05; t = 4.043 in the second week, P <0.01; t = 3.373 in the third week, P <0.01; t = 3.162 in the fourth week , P <0.01). Conclusion The condition fear model can be established by Takeuchi method. Conditional fear and stress can slow the body weight gain and change the behavior and neuroendocrine system.
