目的对ABO正反定型不符的幼儿标本进行血清学鉴定和基因检测分析。方法常规血清学检测,PCR-SSP进行ABO基因初步分型,再使用ABO血型-A亚型基因分型试剂盒进行A亚型分型鉴定。结果血清学结果显示标本正定型为A型(抗A:1+~W),反定为O型(Ac2+,Bc4+);PCR-SSP A亚型分型鉴定结果显示标本基因型为Ax14/O2型。结论疑难血型鉴定有时需要结合血清学和分子生物学方法才能准确定型,此例血清学正反定型不符标本表型为Ax,其基因型为Ax14/O2。
OBJECTIVE: To perform serological identification and genetic testing on young children with non-conforming ABO samples. Methods The routine genotyping and PCR-SSP were used to genotype the ABO gene. Then ABO blood type-A subtype genotyping kit was used to identify the subtype A subtypes. Results The serological results showed that the positive typing was A (anti-A: 1 + ~ W) and the reverse was O (Ac2 +, Bc4 +). The genotyping of PCR-SSP A subtypes showed that the genotype was Ax14 / type. Conclusion Difficult blood group identification sometimes need to be combined with serological and molecular biology methods to accurately shape, this case of serological positive and negative stereotypes inconsistent specimens Ax, its genotype Ax14 / O2.
