目的研究VP16诱导HL-60急性早幼粒白血病细胞程序化死亡。方法用4种不同浓度的VP16诱导HL-60细胞程序化死亡,DNA电泳、电镜及苔盼蓝活细胞拒染试验测定实验结果。结果4种浓度VP16均能诱导HL-60细胞程序化死亡。结论VP16是一种稳定可靠的细胞凋亡诱导剂。苔盼蓝拒染试验不能用于判定细胞是否发生了程序化死亡。DNA电泳是确定细胞程序化死亡的简单可靠的方法。
Objective To investigate the programmed death induced by VP16 in HL-60 acute promyelocytic leukemia cells. Methods The programmed death of HL-60 cells was induced by four different concentrations of VP16. The experimental results were determined by DNA electrophoresis, electron microscopy and trypan blue exclusion test. Results All four concentrations of VP16 could induce apoptosis in HL-60 cells. Conclusion VP16 is a stable and reliable inducer of apoptosis. The Trypan blue exclusion test cannot be used to determine if a programmed cell death has occurred. DNA electrophoresis is a simple and reliable method for determining programmed cell death.
