目的探讨TLR4在BALB/C小鼠肾小管上皮细胞(TEC)的表达和内毒素/脂多糖(LPS)刺激后TLR4的变化,以及对产生肿瘤坏死因子α(TNFα)、白细胞介素-6(IL-6)的影响。方法通过RT-PCR方法检测TLR4 mRNA、TNFαm RNA和IL-6 mRNA在TEC的表达;流式细胞术检测TEC的TLR4膜蛋白水平;ELISA检测培养的TEC上清中TNFα、IL-6的含量。结果正常TEC有TLR4 mRNA表达,但TNFα mRNA、IL-6 mRNA表达甚弱。经LPS刺激后,TEC中的TLR4 mRNA、TNFα mRNA和IL-6 mRNA表达均显著增强,TLR4膜蛋白以及培养上清中的TNFα、IL-6水平亦相应增加。结论TLR4在LPS激活的TEC中表达显著增强,同时TEC分泌的炎症因子TNFα、IL-6的水平也相应上调。
Objective To investigate the changes of TLR4 after TLR4 expression in BALB / c mouse renal tubular epithelial cells (TKIs) and endotoxin / lipopolysaccharide (LPS) stimulation, and the effect of TLR4 on the production of tumor necrosis factor α (TNFα), interleukin- IL-6). Methods The expression of TLR4 mRNA, TNFαmRNA and IL-6 mRNA in TEC was detected by RT-PCR. The TLR4 membrane protein was detected by flow cytometry. The contents of TNFα and IL-6 in the supernatant of TEC were detected by ELISA. Results Normal TEC had TLR4 mRNA expression, but TNFα mRNA and IL-6 mRNA expression were very weak. After LPS stimulation, the expression of TLR4 mRNA, TNFα mRNA and IL-6 mRNA in TEC increased significantly, and the levels of TNFα and IL-6 in TLR4 membrane protein and culture supernatant also increased accordingly. Conclusion The expression of TLR4 in LPS-activated TEC was significantly increased, and the levels of TNFα and IL-6 secreted by TEC were also up-regulated.
