基于ISSR分析的阳春砂分子鉴别

目的利用ISSR分子标记技术鉴别阳春砂不同栽培品种,并分析其亲缘关系。方法利用ISSR-PCR方法对阳春砂栽培品种长果、圆果和“春选”与海南砂进行基因组多态性分析;采用NTSYS-pcversion2.10e软件计算各供试材料间的遗传相似系数,分析其亲缘关系并构建聚类树状图。结果 10条引物共扩增出114条DNA片段,其中49条DNA片段呈现多态性,占总扩增片段的43.0%。长果阳春砂与圆果阳春砂的遗传相似系数最大,高达1.000;长果、圆果阳春砂与海南砂的遗传相似系数最小,为0.577;各材料间的平均遗传相似系数为0.748。UPGMA聚类分析表明,长果阳春砂和圆果阳春砂聚成一支,再与“春选”阳春砂聚成一支,而海南砂独成一支。结论本研究结果鉴别了“春选”和阳春砂的另外两个栽培品种,证明了“春选”与海南砂有着较近的亲缘关系,建立基于ISSR分析的阳春砂遗传分析方法,为阳春砂的品种鉴别和优良品种选育提供了依据。 Objective To identify different cultivars of Populus euphratica by ISSR molecular markers and analyze their genetic relationship. Methods The ISSR-PCR method was used to analyze the genomic polymorphism of the cultivars Longchun, Yuancong, and Spring Spring, and Hainan sand. The genetic similarity coefficients of the tested materials were calculated by NTSYS-pcversion2.10e software , Analyze their kinship and construct cluster dendrogram. Results A total of 114 DNA fragments were amplified with 10 primers. Among them, 49 DNA fragments showed polymorphism, accounting for 43.0% of the total amplified fragment. The genetic similarity coefficient between Changguoyang spring sand and Yuanogangchun spring sand was the highest, reaching as high as 1.000. The genetic similarity coefficient between Changguo and Yuanguo spring sand and Hainan sand was the lowest, which was 0.577. The average genetic similarity coefficient of each material was 0.748. UPGMA cluster analysis showed that the fruit of Changchun Yangchun and Yuanchengyangchun were integrated into one, and then the “Spring election” Yangchun sand was integrated into one, while Hainan sand was the same one. Conclusion The results of this study identified two other cultivars of Spring Spring and Spring Spring, which proved that Spring Spring had a close genetic relationship with Hainan Sand, and established a genetic analysis method based on ISSR , Which provided the basis for the identification of varieties and breeding of good varieties of spring sand.
