目的 :研究香菇多糖对人早幼粒白血病细胞 (HL - 60 )酪氨酸蛋白磷酸化作用的影响。方法 :以不同浓度的药物处理体外培养的HL - 60细胞 ,用放射免疫法测定经过培养 48h的对照组和药物处理组胞浆和膜部分的酪氨酸蛋白激酶 (TPK)和磷酸酪氨酸蛋白磷酸酶 (PTPP)的活力 ,比较酶活力变化情况。结果 :药物对胞浆和膜两部分TPK活力均有一定程度的抑制 ,但抑制作用不明显 (P >0 .0 5) ,而对两部分PTPP活力均有明显的激活作用 (P <0 .0 5或P <0 .0 1 )。结论 :由于上述结果 ,最终导致HL - 60细胞酪氨酸蛋白磷酸化水平下降。提示香菇多糖抗肿瘤作用的分子机制可能与酪酸蛋白磷酸化作用有关
Objective: To study the effect of lentinan on tyrosine phosphorylation of human promyelocytic leukemia cells (HL-60). METHODS: HL-60 cells cultured in vitro were treated with different concentrations of drugs, and tyrosine protein kinase (TPK) and phosphotyrosine were measured by radioimmunoassay in the cytoplasmic and membrane fractions of the control and drug-treated groups after 48 hours of culture. The activity of protein phosphatase (PTPP) compares changes in enzyme activity. RESULTS: The drug had a certain degree of inhibition on TPK activity in the cytoplasm and membrane, but the inhibitory effect was not obvious (P > 0.05), but there was a significant activation effect on the two parts of the PTPP activity (P <0. 0 5 or P <0 .0 1 ). Conclusion: Due to the above results, the tyrosine protein phosphorylation level in HL-60 cells eventually decreased. It is suggested that the molecular mechanism of the anti-tumor effect of lentinan may be related to the phosphorylation of tyrosine protein
