紫外线诱变育种已成为食用菌育种的有效方法之一,但对诱变机理方面的研究甚少。本试验旨在研究紫外线诱变对平菇菌丝长速、生长势和细胞内含物变化的影响,为食用菌紫外线诱变育种提供参考和依据。 一、材料与方法 (一)供试材料 菌株为无孢5号,本校分离。平板培养基为综合马铃薯汁培养基:去皮马铃薯200g,葡萄糖20g,琼脂20g,磷酸二氢钾38,硫酸镁1.5g,维生素B_1 1片,水1000ml,pH值自然。接种后将平板置恒温箱25℃下培养。 (二)照射剂量与方法 试验设1′30″、2′、3′、5′和对照5个处理,每处理重复3次,每重复用18×90mm培养皿平板培养菌丝,每个平板中央接入8mm的菌丝
