用 14种 12个碱基的随机引物对厚皮甜瓜 4个品种及 13个品系进行随机扩增多态性 DNA( RAPD)分析 .结果表明 ,可用其中 6种引物将 4个品种 ( British Queen、Earl’s Favourite、Crenshaw、Pakistan- 981)及Earl’s Favourite的 3个品系 (春系 3号、磐田 2号、夏系 7- 2 )加以鉴别 .在不同引物所扩增的特有或缺少的特异性条带 ( A0 9- 160 0、A0 9- 30 0、A4 1- 2 0 0、A4 2 - 150 0、A4 2 - 90 0、A4 4 - 140、A57- 10 0、A66- 130 0 )可作为鉴定这些品种或品系的分子标记 .在 RAPD分析基础上计算的 4个品种及 13个品系之间的遗传相似性系数 ,与它们的已知来源及彼此间已知的亲缘关系均甚符合
Random amplified polymorphic DNA (RAPD) analysis of four cultivars and 13 strains of melon (Muskmelon) was carried out with 14 12-bp random primers.The results showed that four varieties (British Queen, Earl’s Favorite, Crenshaw, Pakistan-981) and 3 lines of Earl’s Favorite (Spring Line 3, Ibanda 2, Summer Line 7-2). The specific or missing specific bands amplified by different primers (A0 9-160 0, A0 9-30 0, A4 1-200, A4-2-150 0, A4-2- 90 0, A4 4-140, A57-10 0, A66-130 0) can be used as identification Molecular markers of these cultivars or strains The genetic similarity coefficients of the 4 cultivars and 13 cultivars calculated on the basis of RAPD analysis were in good agreement with their known origins and their known relatives
