目的建立检测小鼠血清中刚地弓形虫NTPase-Ⅱ特异性IgGI、gM抗体的间接ELISA法,探讨用于弓形虫感染早期检测的应用价值。方法以刚地弓形虫NTPase-Ⅱ融合蛋白为包被抗原建立间接ELISA法,动态检测刚地弓形虫实验感染鼠血清NTPase-Ⅱ特异性IgGI、gM抗体变化。试验设脑囊虫病、血吸虫病及肺吸虫病病人血清对照。结果小鼠血清中的抗NTPase-Ⅱ抗体IgM于弓形虫感染后第3 d开始出现阳性反应,IgG抗体于感染后第7 d开始出现阳性反应,脑囊虫病、血吸虫病及肺吸虫病病人血清特异性IgG阳性率分别为0、1.92%和0。结论间接ELISA检测弓形虫感染鼠血清NTPase-Ⅱ抗体的敏感性和特异性较高,有望应用于人感染弓形虫的早期检测。
Objective To establish an indirect ELISA for the detection of specific IgGI and gM antibodies against Toxoplasma gondii in mice serum and to explore the value of this method for the early detection of Toxoplasma gondii infection. Methods Indirect ELISA was used to detect T helminthes NTPase-Ⅱ fusion protein as coating antigen to detect the changes of serum NTPase-Ⅱ specific IgGI and gM antibodies in Toxoplasma gondii Mice. Experimental set cysticercosis, schistosomiasis and paragonimiasis patients serum control. Results The anti-NTPase-Ⅱ antibody IgM in sera of mice showed a positive reaction on the 3rd day after infection with Toxoplasma gondii. The IgG antibody showed a positive reaction on the 7th day after infection. Serum specificity of patients with cerebral cysticercosis, schistosomiasis and paragonimiasis The positive rates of IgG were 0, 1.92% and 0 respectively. Conclusion Indirect ELISA detection of Toxoplasma gondii infection with serum NTPase-Ⅱ antibody is highly sensitive and specific, is expected to be applied to early detection of human infection with Toxoplasma gondii.
