目的 :复制一氧化碳 (CO)中毒迟发性脑病大鼠模型。方法 :5 0只Wistar大鼠随机分为 10 0 0ppm组、15 0 0ppm组、16 5 0ppm组、180 0ppm组和对照组共 5组 ,每组 10只。每组中毒箱分别通入浓度为 10 0 0ppm、15 0 0ppm、16 5 0ppm、180 0ppm的CO混合气体 ,对照组通入空气。连续通气 12h。利用迷宫试验评估动物的智力状态。迟发性脑病大鼠诊断标准为中毒 3d后、2 8d内出现昏迷或迷宫实验正确入臂次数少于 3次者。采用TUNEL法和透射电镜等双重方法检测大脑皮质、海马区细胞凋亡情况。结果 :15 0 0ppm组、16 5 0ppm组均有 4只大鼠出现迟发性脑病表现 ,10 0 0ppm组和对照组大鼠未出现迟发性脑病表现 ,180 0ppm组通气 5h有 3只大鼠死亡。两种方法检测凋亡细胞数 ,10 0 0ppm组均少于 5 0 0 % ,对照组均少于 1 6 0 % ,15 0 0ppm组均多于 5 % - 6 5 % ,16 5 0ppm组均多于10 % - 80 %。结论 :建立大鼠CO中毒迟发性脑病模型以CO浓度 15 0 0 - 16 5 0ppm、吸入时间 12h比较适宜。
Objective: To duplicate rat model of delayed encephalopathy caused by carbon monoxide (CO) poisoning. Methods: Fifty Wistar rats were randomly divided into 10 0 ppm group, 150 ppm group, 165 ppm group, 180 ppm group and control group. Each poisoning chamber was introduced into the CO mixed gas at a concentration of 100ppm, 1500ppm, 165Oppm and 180Oppm, respectively. The control group was ventilated with air. Continuous ventilation 12h. Maze test to assess the animal’s mental status. Delayed encephalopathy rat diagnostic criteria for poisoning 3d, within 28 days of coma or maze test correctly into the arm less than 3 times. TUNEL and transmission electron microscopy were used to detect the apoptosis of cerebral cortex and hippocampus. Results: Four hundred and fifty ppm of 15Oppm group showed delayed encephalopathy, delayed onset of encephalopathy was not observed in 100ppm group and control group, and three rats in 180Oppm group Rat died. The number of apoptotic cells in both groups was less than 500% in 100ppm group, less than 160% in control group, more than 5% -65% in 15O0ppm group and 165% in 165ppm group More than 10% - 80%. Conclusion: The establishment of CO poisoning model of delayed encephalopathy in rats with CO concentration of 15 0 0 - 16 50ppm, inhalation time of 12h more appropriate.
