目的探讨经47℃20min短时超高温处理后,胃癌细胞和结肠癌细胞在体外的生长情况,并观察5FU,MMC对肿瘤细胞的细胞毒效应.方法将人胃癌细胞株(Hs746T)、结肠癌细胞株(SW480)和新鲜肿瘤组织细胞,分别经43℃120min或47℃20min热处理后,进行体外细胞毒培养,培养中加入5FU100mg/L,MMC100mg/L,按MTT法检测瘤细胞的生长抑制率,并与未经热处理的肿瘤细胞培养结果相比较.结果热处理可以明显抑制肿瘤细胞的生长,使肿瘤细胞对5FU和MMC的敏感性增加;肿瘤细胞经47℃20min的短时超高温处理后,与43℃120min较长时间热处理后的培养结果相近似;热化疗对新鲜肿瘤组织细胞的生长抑制率与对肿瘤细胞株的生长抑制率相近似.结论47℃20min能达到时间长达120min,温度在43℃的杀伤肿瘤细胞的目的
Objective To investigate the growth of gastric cancer cells and colon cancer cells in vitro after ultra-high temperature treatment at 47°C for 20 min, and observe the cytotoxic effects of 5-FU and MMC on tumor cells. Methods Human gastric cancer cell line (Hs746T), colon cancer cell line (SW480) and fresh tumor cells were heat-treated in vitro at 43°C for 120 minutes or 47°C for 20 minutes. In vitro cytotoxic culture was performed. 5 FU 100 mg/L was added to the culture. MMC 100mg/L, the growth inhibition rate of tumor cells was detected by MTT assay, and compared with the results of tumor cell culture without heat treatment. Results Heat treatment could significantly inhibit the growth of tumor cells and increase the sensitivity of tumor cells to 5FU and MMC. The culture results of tumor cells after heat treatment at 43°C for 120 min after a short-term hyperthermic treatment at 47°C for 20 minutes Similarly, the inhibition rate of thermochemotherapy on the growth of fresh tumor cells is similar to that of tumor cell lines. Conclusion The purpose of killing tumor cells at a temperature of 43°C for a time of up to 120 minutes at 47°C for 20 minutes can be achieved.
