喹乙醇致大鼠肝脏损伤的实验研究

目的探讨喹乙醇(OLA)染毒对实验大鼠肝脏炎症反应及抗氧化酶活性的影响,为OLA致肝损伤机制研究提供理论依据。方法健康雄性SD大鼠32只(SPF级),随机分为对照组、20 mg/kg OLA组、40 mg/kg OLA组和60 mg/kg OLA组,每组实验动物8只。不同剂量OLA染毒组大鼠通过灌胃给不同浓度等体积的OLA溶液;对照组灌胃等体积双蒸水。染毒4周后处死大鼠,留取肝组织做病理观察,应用酶联免疫吸附(ELISA)测定肝组织中白细胞介素-6(IL-6)和肿瘤坏死因子-α(TNF-α)含量,检测血清中丙氨酸转氨酶(ALT)、天冬氨酸转氨酶(AST)、碱性磷酸酶(ALP)和乳酸脱氢酶(LDH)活性,以及肝脏总抗氧化能力(T-AOC)和超氧化物歧化酶(SOD)、谷胱甘肽过氧化物酶(GSH-Px)、过氧化氢酶(CAT)活性。结果随着OLA染毒剂量的增加,血清中ALT、AST、ALP和LDH含量逐渐升高;肝组织中TAOC和SOD、GSH-Px、CAT活性逐渐降低。与对照组比较,OLA组肝组织中TNF-α分别升高了11.68%、12.74%和19.33%,差异有统计学意义(P<0.05);IL-6分别升高了45.99%、70.81%和82.92%,差异有统计学意义(P<0.05);大鼠肝组织TNF-α和IL-6含量与染毒剂量呈正相关(r=0.948,P=0.026和r=0.954,P=0.023)。大鼠肝脏病理观察,OLA组大鼠肝脏汇管区炎性细胞浸润明显。结论 OLA染毒可致大鼠肝组织中发生炎症反应,继而引起肝组织损伤。 Objective To investigate the effects of olaquindox (OLA) on the inflammatory reaction and antioxidase activity in the liver of experimental rats, and provide a theoretical basis for the study on the mechanism of liver injury induced by OLA. Methods Thirty-two healthy male Sprague-Dawley rats were randomly divided into control group, 20 mg / kg OLA group, 40 mg / kg OLA group and 60 mg / kg OLA group, with 8 animals in each group. The rats in different doses of OLA were given different volumes of OLA solution by gavage, while the control group were given the same volume of double distilled water. After 4 weeks of exposure, rats were sacrificed and liver tissues were removed for histopathological examination. Interleukin-6 (IL-6) and tumor necrosis factor-α (TNF-α) were measured by enzyme-linked immunosorbent assay (ELISA) (ALT), aspartate aminotransferase (AST), alkaline phosphatase (ALP) and lactate dehydrogenase (LDH) activity in the serum were measured, and the total liver antioxidant capacity (T-AOC) And superoxide dismutase (SOD), glutathione peroxidase (GSH-Px), catalase (CAT) activity. Results The levels of ALT, AST, ALP and LDH in serum increased gradually with the increase of OLA dose. The activities of TAOC, SOD, GSH-Px and CAT decreased gradually in liver tissue. Compared with the control group, the levels of TNF-α in OLA group increased by 11.68%, 12.74% and 19.33%, respectively, with statistical significance (P <0.05); IL-6 increased by 45.99% and 70.81% 82.92%, respectively. The difference was statistically significant (P <0.05). The content of TNF-α and IL-6 in the liver of the rats was positively correlated with the dosage (r = 0.948, P = 0.026 and r = 0.954, P = 0.023). Pathological observation of rat liver showed that inflammatory cell infiltration in hepatic portal area of ​​OLA group was obvious. Conclusion OLA can cause inflammatory reaction in rat liver tissue, and then cause liver tissue damage.
