目的探讨补中益气汤含药血清对A549/DDP细胞中Bad、NF-κb、caspase-9表达的影响。方法用MTT法筛选最佳浓度的含药血清。用免疫印迹法、real time PCR方法检测A549、A549/DDP细胞各组中Bad、NF-κb、Caspase-9蛋白和mRNA的表达水平。采用SPSS 16.0进行数据处理,P<0.05有统计学意义。结果中剂量含药血清培养48h为最佳浓度含药血清;A549/DDP对照组中Bad、NF-κb蛋白和基因的表达明显高于A549对照组(P<0.05),A549/DDP最佳含药血清组Bad、NF-κb的蛋白和基因表达减少,与A549/DDP对照血清组比较有统计学意义(P<0.05)。A549/DDP对照组中caspase-9蛋白和基因的表达明显低于A549对照组(P<0.05),A549/DDP最佳含药血清组caspase-9的蛋白和基因表达增加,与A549/DDP对照血清组比较有统计学意义(P<0.05)。结论补中益气汤含药血清抑制A549/DDP的作用可能是通过调节Bad、NF-κb、caspase-9蛋白、基因的表达,促进细胞凋亡实现的。
Objective To investigate the effect of Buzhong Yiqi decoction-containing serum on the expression of Bad, NF-κB and caspase-9 in A549 / DDP cells. Methods The optimal concentration of drug-containing serum was screened by MTT method. The expression of Bad, NF-κB, Caspase-9 protein and mRNA in A549, A549 / DDP cells were detected by Western blotting and real time PCR. Data processing using SPSS 16.0, P <0.05 was statistically significant. Results The medium containing serum was cultured for 48h at the best concentration. The expression of Bad and NF-κB protein and gene in A549 / DDP control group was significantly higher than that in A549 control group (P <0.05), A549 / DDP was the best Compared with the A549 / DDP control serum group, the protein and gene expression of Bad and NF-κb in the serum decreased significantly (P <0.05). The expression of caspase-9 protein and gene in A549 / DDP control group was significantly lower than that in A549 control group (P <0.05). The protein and gene expression of caspase-9 in A549 / DDP optimal drug-containing serum group was increased compared with A549 / DDP control group Serum group was statistically significant (P <0.05). Conclusion The suppressive effect of Buzhong Yiqi Decoction containing serum on A549 / DDP may be through regulating the expression of Bad, NF-κB, caspase-9 protein and gene and promoting cell apoptosis.
