目的探讨维吾尔族和汉族女性乳腺癌与KIAA1199表达调控的关系。方法收集维吾尔族和汉族女性乳腺纤维腺瘤、乳腺癌患者的石蜡包埋组织标本150例(维吾尔族68例,汉族82例),另选取新鲜组织标本58例(维吾尔族21例,汉族37例),采用免疫组织化学和逆转录聚合酶链反应(RT-PCR)法检测KIAA1199蛋白和mRNA表达水平。结果乳腺纤维腺瘤细胞中KIAA1199蛋白表达上调率为31.9%,乳腺癌细胞中KIAA1199蛋白表达上调率为76.7%,差异有统计学意义(P<0.01),维吾尔族、汉族女性乳腺癌KIAA1199表达上调率的变化趋势具有共性(维吾尔族2=12.30,P<0.01;汉族2=15.19,P<0.01),其族群差异无统计学意义(P>0.05)。KIAA1199基因的转录表达水平在乳腺癌组织中明显上调,其mRNA相对含量为0.967±0.281,乳腺纤维腺瘤组织中KIAA1199 mRNA含量为0.475±0.176,差异有统计学意义(P<0.01)。结论 KIAA1199表达上调伴随着乳腺病变进程,可能成为乳腺癌分子预警指标。
Objective To investigate the relationship between the expression of KIAA1199 and Uighur and Han female breast cancer. Methods 150 cases of UW and Han women with breast fibroadenoma and breast cancer were enrolled in this study. Among them, 150 cases were Uygur and 82 Han cases, and 58 cases were fresh tissue samples (21 Uygur and 37 Han cases) ), Immunohistochemistry and reverse transcription polymerase chain reaction (RT-PCR) assay KIAA1199 protein and mRNA expression levels. Results The up-regulation rate of KIAA1199 protein was 31.9% in breast fibroadenoma cells and 76.7% in breast cancer cells (P <0.01) (Uygur 2 = 12.30, P <0.01; Han 2 = 15.19, P <0.01). There was no significant difference in ethnicity between the two groups (P> 0.05). The transcriptional level of KIAA1199 gene was significantly up-regulated in breast cancer tissues, the relative content of mRNA was 0.967 ± 0.281, and the content of KIAA1199 mRNA in breast fibroadenoma was 0.475 ± 0.176 (P <0.01). Conclusion The up-regulation of KIAA1199 is associated with the progression of breast lesions and may be an early warning marker for breast cancer.
