本研究采用免疫细胞化学PAP方法,对培养4天的E_8鸡胚前脑5-羟色胺(5-HT)免疫反应神经细胞的表达进行研究。发现生长在基板蛋白(LN)支持物上的前脑细胞,98%为神经特异性烯醇酶(NSE)免疫反应阳性细胞,证明为神经细胞;其中部分神经细胞显示5-HT免疫反应强阳性,约占培养神经细胞的10.4%,大多数细胞为多突或双极的形态学特征。为了要了解其余2%的NSE免疫反应阴性细胞是否为胶质细胞,我们以胶质纤维酸性蛋白(GFAP)抗体做胶质细胞特异性染色,阳性细胞为2%,与上述结果一致。
In this study, immunocytochemical PAP method was used to study the expression of 5-hydroxytryptamine (5-HT) immunoreactive nerve cells in E_8 chicken embryo cultured for 4 days. It was found that 98% of forebrain cells growing on substrate protein (LN) support were neuron specific enolase (NSE) immunoreactive cells, which were proved to be neural cells. Some neurons showed strong positive 5-HT immune response , Accounting for about 10.4% of cultured nerve cells, most of the cells are multi-sudden or bipolar morphological features. To see if the remaining 2% of NSE immunoreactive negative cells were glial cells, we performed glial cell-specific staining with glial fibrillary acidic protein (GFAP) antibody, with 2% positive cells, consistent with the above results.
