目的:评价益母散对孕鼠分娩能力的促进作用,并初步探讨其作用机制。创新点:首次在临近分娩小鼠中证明新中兽医药处方益母散可明显促进孕鼠分娩能力,且此作用与催产素和子宫平滑肌缝隙连接蛋白43(Cx43)的高表达相关。方法:将妊娠小鼠分为对照组和益母散低、中、高剂量组,产前3天持续灌胃,统计孕鼠分娩产程和死胎率。采用离体平滑肌张力试验测定子宫平滑肌张力,用酶联免疫吸附测定(ELISA)方法检测血清中催产素和前列腺素E2(PGE2)含量,用蛋白免疫印迹(Western blotting)及免疫组织化学等方法检测小鼠子宫平滑肌中Cx43含量。结论:益母散灌服处理能够使小鼠分娩产程缩短和死胎率降低(图1),并增强分娩小鼠子宫平滑肌张力(图2)。ELISA实验结果显示,益母散灌服处理使分娩小鼠血清催产素水平显著提高(图3)。免疫组化和免疫印迹实验结果显示,益母散灌服处理使分娩小鼠子宫平滑肌Cx43含量显著提高(图4和5)。综上所述,益母散具有提高孕鼠分娩能力的作用,该作用与催产素水平和子宫平滑肌Cx43的高表达相关。
Objective: To evaluate the effect of Yimousan on the delivery ability of pregnant rats and its mechanism of action. Innovative point: For the first time in farms close to birth to prove that the new Chinese medicine prescription Yimusan can significantly promote the delivery ability of pregnant rats, and this role and oxytocin and uterine smooth muscle connexin 43 (Cx43) high expression. Methods: The pregnant mice were divided into control group and Yusan San low, medium and high dose groups. The rats were given gavage 3 days prenatally, the labor process and the stillbirth rate of pregnant mice were counted. Uterine smooth muscle tension was measured by in vitro smooth muscle tension test, and serum levels of oxytocin and prostaglandin E2 (PGE2) were detected by enzyme-linked immunosorbent assay (ELISA). The protein levels of PGE2 were detected by Western blotting and immunohistochemistry Cx43 content in mouse uterine smooth muscle. Conclusion: Yusumosan can shorten the delivery process and the stillbirth rate of mice (Figure 1) and enhance the uterine smooth muscle tension of the delivery mice (Figure 2). ELISA test results show that Yimousi Sanfu treatment so that the delivery of serum oxytocin levels were significantly increased (Figure 3). The results of immunohistochemistry and Western blotting showed that the treatment of Yimuzhuo San was significantly improved Cx43 content in uterine smooth muscle of farrowing mice (Figures 4 and 5). In conclusion, Yimuzan Powder has the effect of improving the delivery ability of pregnant mice, which is related to the high expression of oxytocin and uterine smooth muscle Cx43.
