目的:探讨肝细胞癌(HCC)组织中ABCG2和Jagged1的表达状况,分析两者与肝癌临床病理学参数的相关性及临床意义。方法:分别应用免疫组化SP法和原位杂交的方法检测150例HCC、50例肝硬化和10例正常肝组织中ABCG2和Jag-ged1蛋白及mRNA的表达情况。结果:ABCG2蛋白在HCC、肝硬化和正常肝组织的阳性表达率分别为78.67%(118/150)﹑40.00%(20/50)和30.00%(3/10),差异有统计学意义,χ2=31.98,P<0.05。ABCG2mRNA在三种组织中的阳性表达率分别为69.33%(104/150)、36.00(18/50)和20.00%(2/10),差异有统计学意义,χ2=23.85,P<0.05。ABCG2蛋白在HCC组织中的表达与有无转移有关,χ2=10.93,P<0.05。Jagged1蛋白在HCC﹑肝硬化﹑正常肝组织中的阳性表达率分别为71.33%(107/150)、52.00%(26/50)和20.00%(2/10),差异有统计学意义,χ2=15.07,P<0.05。Jagged1mRNA在三种组织中的阳性表达率分别为60.67%(91/150)、36.00%(18/50)和20.00%(2/10)差异有统计学意义,χ2=13.71,P<0.05。Jagged1蛋白表达与年龄(χ2=7.94,P<0.05)、组织学分级(χ2=14.79,P<0.05)及HBsAg(χ2=4.83,P<0.05)有关。ABCG2蛋白的表达与Jagged1蛋白表达呈正相关,r=0.458,P<0.05。结论:ABCG2和Jagged1可能成为判断HCC预后和分子生物学行为的重要指标。
OBJECTIVE: To investigate the expression of ABCG2 and Jagged1 in hepatocellular carcinoma (HCC) tissues and to analyze their correlation with the clinicopathological parameters of hepatocellular carcinoma and its clinical significance. Methods: The expressions of ABCG2 and Jag-ged1 protein in 150 cases of HCC, 50 cases of cirrhosis and 10 cases of normal liver tissue were detected by immunohistochemical SP method and in situ hybridization respectively. Results: The positive expression rates of ABCG2 protein in HCC, cirrhosis and normal liver tissues were 78.67% (118/150), 40.00% (20/50) and 30.00% (3/10), respectively, with significant difference (χ2 = 31.98, P <0.05. The positive rates of ABCG2 mRNA in the three tissues were 69.33% (104/150), 36.00 (18/50) and 20.00% (2/10), respectively. The difference was statistically significant (χ2 = 23.85, P <0.05). The expression of ABCG2 protein in HCC tissue correlated with or without metastasis, χ2 = 10.93, P <0.05. The positive rates of Jagged1 protein in HCC, cirrhosis and normal liver tissues were 71.33% (107/150), 52.00% (26/50) and 20.00% (2/10), respectively, the difference was statistically significant (χ2 = 15.07, P <0.05. The positive rates of Jagged1 mRNA in the three tissues were 60.67% (91/150), 36.00% (18/50) and 20.00% (2/10), respectively. The difference was statistically significant (χ2 = 13.71, P <0.05). Jagged1 protein expression was related to age (χ2 = 7.94, P <0.05), histological grade (χ2 = 14.79, P <0.05) and HBsAg (χ2 = 4.83, P <0.05). The expression of ABCG2 protein was positively correlated with the expression of Jagged1 protein (r = 0.458, P <0.05). Conclusion: ABCG2 and Jagged1 may be important indicators to judge the prognosis and molecular biological behavior of HCC.
