目的探讨小剂量线粒体毒素3-硝基丙酸(3-NPA)预处理对红藻氨酸(KA)致痫大鼠海马神经细胞凋亡和p53蛋白表达的影响。方法大鼠腹腔注射20mg/kg3-NPA(4mg/mL)或生理盐水后24h制作大鼠癫痫模型及对照模型,7d后分别用原位末端标记(TUNEL)法、免疫组织化学方法观察小剂量3-NPA预处理对KA致痫大鼠海马CA1区神经细胞凋亡和P53蛋白表达的影响。结果3-NPA预处理组较对照组CA1区神经细胞凋亡减少,p53蛋白表达减弱。结论小剂量3-NPA预处理可以对KA致痫大鼠海马神经细胞凋亡和p53蛋白表达有抑制作用,3-NPA预处理可能对KA致痫大鼠海马细胞凋亡具有一定保护作用。
Objective To investigate the effect of 3-NPA pretreatment on neuronal apoptosis and p53 protein expression in hippocampus induced by kainic acid (KA) in rats. Methods Rats were injected intraperitoneally with 20mg / kg 3-NPA (4mg / mL) or normal saline for 24 hours to make epilepsy rat model and control model. After 7 days, TUNEL method and immunohistochemical method were used to observe the effects of low dose 3 Effect of N - PA Pretreatment on Nerve Cell Apoptosis and P53 Protein Expression in Hippocampal CA1 Area of KA - induced Epileptic Rats. Results Compared with the control group, the apoptosis of neurons in 3-NPA preconditioning group decreased and the expression of p53 protein decreased. Conclusion Pretreatment with low-dose 3-NPA can inhibit the apoptosis of hippocampal neurons and the expression of p53 protein in KA-induced epileptic rats. Pretreatment with 3-NPA may protect hippocampal cells from KA-induced seizures.
