目的:观察清肺化痰通络方对肺间质纤维化大鼠肺内胶原纤维沉积和转化生长因子-β1(TGF-β1)表达的影响。方法:Wistar雌性大鼠72只,随机分为对照组、模型组、中药组。模型组和中药组大鼠气管内一次性滴注博莱霉素A5(BLMA5),对照组大鼠气管内一次性滴注等体积的生理盐水。造模后2h开始给药,模型组和对照组灌服生理盐水,中药组灌服清肺化痰通络方药物,每天一次。各组动物分别于第3、7、14、28天各处死6只,取肺组织进行HE和Masson染色,观察肺内胶原的沉积部位和数量;同时进行免疫组化染色,结合图像分析来检测各组肺组织中TGF-β1的表达水平。结果:对照组肺内结构正常,胶原沉积不明显;模型组可见支气管黏膜下、血管壁及其周围、肺间质、肺泡壁有大量胶原沉积;中药组胶原沉积明显轻于模型组。第14天模型组、中药组TGF-β1在肺内的表达呈强阳性,高于对照组,差异有显著性意义(P<0.01)。中药组TGF-β1表达数量少于同一时段模型组,差异有显著性意义(P<0.05)。结论:清肺化痰通络方可以抑制肺间质纤维化大鼠肺组织内胶原的沉积,可以减缓肺纤维化的形成,其作用机制可能是通过抑制TGF-β1的表达来实现。
Objective: To observe the effects of Qingfei Huatan Tongluo Prescription on the expression of collagen fibers and transforming growth factor-β1 (TGF-β1) in lungs of rats with pulmonary interstitial fibrosis. Methods: Seventy-two Wistar female rats were randomly divided into control group, model group and TCM group. The rats in the model group and the Chinese herb group were given a single instillation of bleomycin A5 (BLMA5) in the trachea, and the rats in the control group were given a single intravenous infusion of equal volume of normal saline. 2h after model administration, the model group and the control group were fed with saline, and the Chinese medicine group was given Qingfei Huatan Tongluo recipe once a day. Each group of animals were sacrificed on the 3rd, 7th, 14th and 28th day, respectively. Six rabbits were sacrificed on the 3rd, 7th, 14th and 28th days respectively. HE and Masson stained lungs were used to observe the deposition site and number of collagen in the lungs. Meanwhile, immunohistochemistry and image analysis The expression of TGF-β1 in each group of lung tissue. Results: In the control group, the lung structure was normal and the collagen deposition was not obvious. In the model group, there was a large amount of collagen deposition in the bronchial submucosa, blood vessel wall and its surroundings, interstitial lung and alveolar wall. The collagen deposition in the TCM group was lighter than that in the model group. On the 14th day, the expression of TGF-β1 in the model group and the traditional Chinese medicine group was strongly positive in the lung, which was higher than that in the control group (P <0.01). The number of TGF-β1 expression in the TCM group was less than that in the model group at the same time point, the difference was significant (P <0.05). Conclusion: Qingfei Huatan Tongluo Prescription can inhibit the collagen deposition in the lung tissue of rats with pulmonary interstitial fibrosis and slow down the formation of pulmonary fibrosis. Its mechanism may be through inhibiting the expression of TGF-β1.
