目的:探讨丹红注射液对脑缺血再灌注后脑细胞凋亡诱导因子(AIF)表达以及神经元凋亡情况的影响。方法:将20只大鼠随机分为假手术组2只,脑缺血再灌注组9只,丹红注射液组9只。建立脑缺血再灌注动物模型,并于再灌注24h、48h、72h时分别检测脑缺血再灌注组和丹红注射液组大鼠的AIF阳性和神经细胞凋亡阳性情况。结果:假手术组大鼠术后24h脑组织切片AIF检测未见阳性,脑缺血再灌注组术后24hAIF阳性数极显著高于丹红注射液组(P<0.01),术后48h显著高于丹红注射液组(P<0.05),术后72h两组无显著差异(P>0.05)。假手术组神经元凋亡检测未见阳性,脑缺血再灌注组24h凋亡量明显高于丹红注射液组(P<0.05),48h和72h两组无显著差异(P>0.05)。结论:丹红注射液有助于减少脑缺血再灌注后的神经元凋亡,提高对缺血脑细胞的保护作用,拮抗神经元凋亡的最佳时机在灌注的24h内。
Objective: To investigate the effect of Danhong injection on the expression of apoptosis-inducing factor (AIF) and neuronal apoptosis after cerebral ischemia-reperfusion. Methods: Twenty rats were randomly divided into sham operation group (n = 2), cerebral ischemia reperfusion group (n = 9) and Danhong injection group (n = 9). The animal model of cerebral ischemia-reperfusion was established. The positive AIF-positive and neuronal apoptosis in cerebral ischemia-reperfusion group and Danhong injection group were detected at 24h, 48h, 72h after reperfusion. Results: AIF expression in brain tissue of sham operation group was not found positive 24 hours after operation. The positive number of 24h AIF in cerebral ischemia-reperfusion group was significantly higher than that in Danhong injection group (P <0.01), and significantly higher than that in Dan Red injection group (P <0.05), but there was no significant difference between the two groups at 72h after operation (P> 0.05). The apoptosis of neurons in sham operation group was not detected. The apoptosis of cerebral ischemia-reperfusion group was significantly higher than that of Danhong injection group (P <0.05), but there was no significant difference between the two groups at 48h and 72h (P> 0.05). Conclusion: Danhong injection can reduce neuronal apoptosis after cerebral ischemia and reperfusion, improve the protective effect on ischemic brain cells, and antagonize the best timing of neuronal apoptosis within 24h of perfusion.
