目的:通过检测胰腺癌、慢性胰腺炎及正常胰腺标本中的平均微血管密度(MVD)及血管内皮细胞生长因子(VEGF)表达水平,了解胰腺癌肿瘤血管生成情况。方法:胰腺癌标本26例,慢性胰腺炎标本7例,癌旁正常胰腺组织26例(采用胰腺癌标本中无肿瘤浸润的组织),所有标本均行免疫组化染色。MVD检测为在200倍显微镜下计数5个视野最高数目的微血管,以其平均数表示。VEGF的定量标准为VEGF染色强度与阳性细胞百分比分值的乘积。统计分析采用t检验和二元变量相关分析法。结果:VEGF在胰腺癌、慢性胰腺炎及癌旁正常胰腺组织中均有表达。胰腺癌组织的MVD值及VEGF表达水平均明显高于慢性胰腺炎及癌旁正常胰腺组织(P<0.01)。26例胰腺癌组织MVD值及VEGF表达水平在统计学上有相关性(Spearman法r=0.862,P<0.000;Pearson法r=0.855,P<0.000)。结论:胰腺癌的微血管生成水平高于慢性胰腺炎及癌旁正常胰腺组织;胰腺癌组织内MVD值及VEGF表达水平在统计学上有相关性。
OBJECTIVE: To understand the angiogenesis of pancreatic cancer by detecting the average microvessel density (MVD) and the expression of vascular endothelial growth factor (VEGF) in pancreatic cancer, chronic pancreatitis and normal pancreas. Methods: Twenty-six pancreatic cancer specimens, seven chronic pancreatitis specimens and 26 normal pancreatic tissues (using no tumor-infiltrating tissue in pancreatic cancer specimens), all specimens were examined by immunohistochemistry. MVD detection is counting of the highest number of capillaries in five fields under a 200-fold microscope, expressed as an average number thereof. The quantitative standard for VEGF is the product of the intensity of VEGF staining and the percentage of positive cells. Statistical analysis using t test and binary variable correlation analysis. Results: VEGF was expressed in pancreatic cancer, chronic pancreatitis and adjacent normal pancreatic tissue. MVD and VEGF expression in pancreatic cancer tissues were significantly higher than those in chronic pancreatitis and adjacent normal pancreatic tissues (P <0.01). MVD and VEGF expression in 26 cases of pancreatic cancer were statistically significant (Spearman’s r = 0.862, P <0.000; Pearson’s r = 0.855, P <0.000). Conclusions: The microvessel formation level of pancreatic cancer is higher than that of chronic pancreatitis and adjacent normal pancreatic tissues. There is a statistically significant correlation between MVD and VEGF expression in pancreatic cancer.
