目的:建立枸杞子中甜菜碱含量的HPLC-ELSD测定方法。方法:采用Hypersil NH2色谱柱(250 mm×4.6 mm,5μm),流动相为甲醇-四氢呋喃-0.2%三氟乙酸(30:60:10),流速为1.0 mL.min-1,柱温为25℃。结果:甜菜碱进样量为1.28~12.8μg时,其与色谱峰面积具有良好的线性关系(r=0.999 2);平均加样回收率为98.60%,RSD为2.8%(n=6);测得样品中甜菜碱含量为0.528 1~1.205 4 mg.g-1。结论:该HPLC-ELSD法简便,准确度高,可用于枸杞子的质量控制。
Objective: To establish a method for the determination of betaine in Fructus Lycii by HPLC-ELSD. Methods: Hypersil NH2 column (250 mm × 4.6 mm, 5 μm) was used with the mobile phase consisted of methanol-tetrahydrofuran-0.2% trifluoroacetic acid (30:60:10) at a flow rate of 1.0 mL · min- ℃. Results: The linear regression was obtained when the injection volume of betaine was between 1.28 and 12.8μg (r = 0.999 2). The average recovery was 98.60% and the RSD was 2.8% (n = 6). Measured in the sample betaine content of 0.528 1 ~ 1.205 4 mg.g-1. Conclusion: The HPLC-ELSD method is simple, accurate and can be used for the quality control of Chinese wolfberry fruit.
