目的 采用一种新的多聚阳离子化合物——多聚乙酰亚胺 (polyethylenim ine,PEI) ,作为将外源性基因转入骨骼肌细胞的介导物。 方法 骨骼肌细胞取自新生 SD大鼠 ,经胰酶消化后培养 ,将 L ac Z基因 DNA - PEI复合物导入骨骼肌细胞内 ,最后用 X- gal组织化学染色检测已转染的细胞。 结果 PEI可作为介导剂使 L ac Z基因在培养的骨骼肌细胞中表达 ,其表达率可达到 30 %。 结论 PEI在真核细胞中实施基因转移是一种有效方法 ,为基因治疗、研究提供了一种新的技术途径
Aim To use polyethylenimine (PEI), a new polycationic compound, as a mediator of exogenous gene transfer into skeletal muscle cells. Methods Skeletal muscle cells were isolated from newborn SD rats and were digested with trypsin. The L ac Z gene DNA - PEI complex was introduced into skeletal muscle cells. The transfected cells were detected by X - gal histochemical staining. Results PEI could be used as a mediator to express L ac Z gene in cultured skeletal muscle cells with the expression rate of up to 30%. Conclusion PEI is an effective method for gene transfer in eukaryotic cells, which provides a new technical approach for gene therapy and research
