目的:研究自发性2型糖尿病(T2DM)大鼠ZDF(Zucker diabetic fatty)(fa/fa)与ZDF(fa/+)及经糖耐康干预后,ZDF(fa/fa)肠道共生细菌菌群结构的变化。方法:将6只雄性ZDF(fa/+)大鼠设为正常组,雄性ZDF(fa/fa)大鼠18只(不同日随机血糖均值≥11.1 mmol·L~(-1))设为T2DM成模组。根据体重及随机血糖(RBG)随机分为3组,分别为模型组(生理盐水10 mL·kg~(-1)),糖耐康高、低剂量(糖耐康3.24,0.81 g·kg~(-1))组,每组6只。给药6周后取材,腹主动脉取血检测大鼠空腹血糖(FPG)及空腹血清胰岛素(FINS);收集大鼠肠道内粪便,行逆转录-聚合酶链式反应(RT-PCR),通过扩增子焦磷酸测序分析16S rRNA V4区菌群的改变。结果:与模型组比较,糖耐康高、低剂量组大鼠FPG,FINS有显著改善(P<0.01)。各组大鼠在菌群丰度多样性及结构组成上有显著差异。在科及以下水平上,与正常组比较,模型组乳酸杆菌明显升高(P<0.05);与模型组比较,糖耐康高、低剂量组乳酸杆菌丰度有所下降,但无统计学差异,普雷沃氏菌丰度显著升高(P<0.01),且普雷沃氏菌属与ZDF大鼠FINS呈正相关。结论:ZDF(fa/fa)与ZDF(fa/+)大鼠在肠道菌群结构上有很大差异,糖耐康对ZDF大鼠血糖的改善作用可能是通过影响ZDF大鼠肠道中乳酸杆菌、普雷沃氏菌的丰度实现。
Objective: To investigate the effects of ZDF (fa / fa) and ZDF (fa / +) and ZDF (fa / fa) intestinal commensal bacteria on the pathogenesis of spontaneous type 2 diabetes mellitus Group structure changes. Methods: Six male ZDF (fa / +) rats were set as normal group, and 18 male ZDF (fa / fa) rats (mean blood glucose level ≥11.1 mmol·L -1 on different days) Into the module. The feces in the intestine of the rats was collected and RT-PCR was performed. The 16S rRNA V4 flora was analyzed by amplicon pyrosequencing. Results: Compared with the model group, FPG and FINS in high and low dose Glucose-treated rats were significantly improved (P <0.01). Each group of rats in the diversity of abundance and structural composition of significant differences. Compared with the normal group, Lactobacillus in the model group was significantly higher than that in the normal group (P <0.05). Compared with the model group, the abundance of lactobacilli decreased in the high and low dose Glucose-resistant groups, but there was no statistical difference (P <0.01), and there was a positive correlation between Prevotella genus and FINS in ZDF rats. Conclusion: ZDF (fa / fa) and ZDF (fa / +) rats have great differences in intestinal microflora. The improvement of glucose in ZDF rats may be mediated by lactate Bacillus, Prevotella abundance achieved.
