目的:研究弯柄灵芝多糖加速溶剂萃取技术(ASE)提取工艺。方法:以弯柄灵芝多糖含量为指标,蒽酮-浓硫酸法测定,并结合加速溶剂萃取法,采用L9(3~3)正交试验对弯柄灵芝多糖提取工艺优化设计。结果:加速溶剂萃取技术最佳提取工艺条件为提取温度180℃,提取时间12 min,静态循环提取次数2次,提取温度对其多糖提取率具有极显著性影响(P<0.01)。在最优提取条件下,弯柄灵芝多糖提取率为(12.66±1.05)%。结论:该工艺与传统水提醇沉提取法相比,具有操作简便、缩短提取时间、节能耗、多糖得率高等优点,可用于弯柄灵芝多糖的提取生产,为弯柄灵芝产品的开发提供了有益的参考。
Objective: To study the extraction technology of polysaccharides from Acanthopanax glabra by accelerated solvent extraction (ASE). Methods: Optimizing the extraction process of Ganoderma lucidum polysaccharides by L9 (3 ~ 3) orthogonal test was carried out with anthrone - concentrated sulfuric acid method and accelerated solvent extraction method. Results: The optimum conditions of extraction by accelerated solvent extraction were extraction temperature 180 ℃, extraction time 12 min and number of static cycles 2 times. The extraction temperature had a significant effect on the extraction rate of polysaccharide (P <0.01). Under optimal extraction conditions, the extraction rate of Ganoderma lucidum polysaccharides was (12.66 ± 1.05)%. Conclusion: Compared with the traditional water extraction alcohol sedimentation extraction method, this method has the advantages of simple operation, shortening extraction time, energy saving, high yield of polysaccharide, etc. It can be used in the extraction of polysaccharides from Curcuma longan Useful reference.
