目的研究甜菜碱对异丙肾上腺素诱导大鼠急性缺血性心肌损伤炎症因子表达的抑制作用。方法用甜菜碱(100、200、400 mg·kg-1·d-1)灌胃大鼠40 d,第39、40天于皮下注射85 mg·kg-1·d-1异丙肾上腺素;取大鼠左心室的心肌组织,采用蛋白芯片技术再结合酶联免疫检测方法,研究甜菜碱对急性缺血心肌炎症因子表达的影响。结果甜菜碱对心肌急性缺血性损伤具有保护作用。与正常组比较,试验组中急性缺血心肌炎症因子β-NGF、TIMP-1、ICAM-1、MCP-1的表达增加,IL-1β、IL-4、VEGF的表达减少;给予大鼠甜菜碱(200、400 mg·kg-1·d-1),对缺血心肌MCP-1和ICAM-1的高表达具有抑制作用。结论甜菜碱可通过抑制心肌MCP-1和ICAM-1的表达对急性心肌缺血性损伤具有保护作用。
Objective To study the inhibitory effect of betaine on isoproterenol-induced inflammatory factor expression in acute ischemic myocardial injury in rats. Methods Rats were gavaged with betaine (100, 200, 400 mg · kg-1 · d-1) for 40 days. On the 39th and 40th day, rats were injected subcutaneously with 85 mg · kg-1 · d-1 isoproterenol. The myocardial tissue of left ventricle of rats was taken and the effect of betaine on the expression of inflammatory cytokines in acute ischemic myocardium was studied by protein chip technique combined with enzyme-linked immunosorbent assay. Results Betaine has a protective effect on myocardial acute ischemic injury. Compared with the normal group, the expression of inflammatory factor β-NGF, TIMP-1, ICAM-1 and MCP-1 in acute ischemic myocardium increased and the expression of IL-1β, IL-4 and VEGF decreased in the experimental group; Alkaloids (200, 400 mg · kg-1 · d-1) inhibited the high expression of MCP-1 and ICAM-1 in ischemic myocardium. CONCLUSION Betaine can protect acute myocardial ischemic injury by inhibiting the expression of MCP-1 and ICAM-1.
