背景:有研究表明CpG寡脱氧核苷酸可增强外周血单个核细胞的功能,但对1型糖尿病患者的影响至今少有报道。目的:观察CpG寡脱氧核苷酸对1型糖尿患者患者与健康志愿者外周血单个核细胞γ干扰素、白细胞介素12,10表达的影响。方法:将1型糖尿病患者与健康志愿者的外周血单个核细胞根据刺激物不同分为空白对照组、CpG寡脱氧核苷酸组。用RT-PCR法检测外周血单个核细胞γ干扰素、白细胞介素12mRNA和白细胞介素10mRNA的表达。结果与结论:1型糖尿患者γ干扰素和白细胞介素12mRNA的表达明显低于健康志愿者(P<0.01)。1型糖尿患者与健康志愿者外周血单个核细胞经CpG寡脱氧核苷酸组刺激后,γ干扰素和白细胞介素12mRNA的表达增高(P<0.01),白细胞介素10mRNA的表达无差异(P>0.05)。结果提示,CpG寡脱氧核苷酸可促进1型糖尿患者外周血单个核细胞表达γ干扰素和白细胞介素12。
BACKGROUND: Studies have shown that CpG oligodeoxynucleotides enhance the function of peripheral blood mononuclear cells, but so far the impact on patients with type 1 diabetes has rarely been reported. Objective: To observe the effect of CpG oligodeoxynucleotides on the expression of interferon-γ and interleukin-12 in peripheral blood mononuclear cells in patients with type 1 diabetes mellitus and healthy volunteers. Methods: Peripheral blood mononuclear cells from patients with type 1 diabetes and healthy volunteers were divided into blank control group and CpG oligodeoxynucleotide group according to different stimuli. The expression of interferon-γ, interleukin-12 mRNA and interleukin-10 mRNA in peripheral blood mononuclear cells were detected by RT-PCR. RESULTS AND CONCLUSION: The expression of IFN-γ and IL-12 mRNA in type 1 diabetes patients was significantly lower than that in healthy volunteers (P <0.01). The expression of IFN - γ and IL - 12 mRNA was increased in PBMCs of type 1 diabetic patients and healthy volunteers after stimulation by CpG oligodeoxynucleotides (P <0.01), but there was no difference in the expression of IL - 10 mRNA P> 0.05). The results suggest that CpG oligodeoxynucleotides can promote the expression of IFN-γ and IL-12 in peripheral blood mononuclear cells of type 1 diabetic patients.
