就应用强酸性阳离子交换树脂(B)及强碱性阴离子交换树脂(G)分离提纯发酵液中L-谷氨酰胺(L-Gln)的工艺条件进行了研究,通过单因子及正交试验确定离子交换分离法分离L-Gln的较佳条件为:强酸性阳离子交换树脂分离L-Gln的上柱液pH为3.5,洗脱剂氨水的浓度为0.03mol/L,洗脱速度为树脂体积的0.65%;强碱性阴离子交换树脂分离L-Gln的上柱液的pH为8.0,洗脱剂盐酸的浓度为0.05mol/L,洗脱速度为树脂体积的1.15%;使用上述条件提取L-Gln的得率为30%左右。
The conditions for the separation and purification of L-glutamine (L-Gln) from the fermentation broth using strong acid cation exchange resin (B) and strong basic anion exchange resin (G) were studied by single factor and orthogonal test The optimum conditions for the separation of L-Gln by ion-exchange separation are as follows: the pH of the upper column for the separation of L-Gln from the strongly acidic cation exchange resin is 3.5, the concentration of the eluent ammonia is 0.03 mol / L, the elution rate is 0.65% by volume of the resin; the pH of the supernatant liquid separating the L-Gln from the strongly basic anion exchange resin was 8.0, the concentration of the eluant hydrochloric acid was 0.05 mol / L, and the elution rate was 1 .15%.
