目的观察银杏酮酯(GBE50)对抑郁模型大鼠行为学的影响,并检测各组大鼠血清皮质酮浓度,探讨GBE50抗抑郁的效果。方法采用国内外实验室普遍应用的慢性不可预见性应激抑郁模型。将60只雄性SD大鼠随机分为6组,包括正常对照组,模型组,西药(氟西汀)组,中药(GBE50)高、中、低剂量组。经21天应激结合同时给药,对各实验组进行强迫游泳实验测试并检测各组血清皮质酮含量。结果与正常对照组比较,模型组大鼠挣扎时间明显缩短(P<0.01),不动时间则显著延长(P<0.01);与模型组比较,中药高、中剂量组可显著延长大鼠挣扎时间(P<0.01),西药组和中药高、中剂量组可明显缩短大鼠不动时间(P<0.01)。与正常对照组比较,模型组大鼠血清皮质酮浓度明显增加(P<0.01);与模型组比较,氟西汀组及中药高、中剂量组大鼠的血清皮质酮浓度均显著下降(P<0.01)。结论 GBE50可显著缓解模型大鼠抑郁样行为,提示GBE50具有一定治疗抑郁症作用。
Objective To observe the effect of GBE50 on the behavior of depression model rats and to detect the serum corticosterone concentration of rats in each group and to explore the antidepressant effect of GBE50. Methods Chronic unpredictable stress depression models commonly used in laboratories at home and abroad were used. Sixty male Sprague-Dawley rats were randomly divided into 6 groups, including normal control group, model group, western medicine (fluoxetine) group and traditional Chinese medicine (GBE50) high, medium and low dose groups. After 21 days of stress combined with simultaneous administration, forced swimming test was conducted on each experimental group and serum corticosterone levels of each group were measured. Results Compared with the normal control group, the struggling time of the model group was significantly shorter (P <0.01) and the immobility time was significantly longer (P <0.01). Compared with the model group, the high and medium dose groups of Chinese medicine significantly prolonged the rat struggles (P <0.01), Western medicine group and traditional Chinese medicine high and medium dose group can obviously shorten the immobility time of rats (P <0.01). Compared with the normal control group, the concentration of serum corticosterone in the model group was significantly increased (P <0.01). Compared with the model group, the serum corticosterone concentrations in the fluoxetine group and the high-dose and middle-dose groups of rats were significantly decreased (P <0.01). Conclusion GBE50 can significantly alleviate the depression-like behavior of model rats, suggesting that GBE50 has a certain role in the treatment of depression.
