目的 考察天麻素对由氯化钾诱导的神经细胞释放谷氨酸的影响。方法 通过高效液相 (HPLC)测定人神经母细胞瘤SH SY5Y细胞系培养液中谷氨酸的含量。结果 当细胞外液中有钙离子时 ,随着氯化钾浓度的增加 (10~ 30mmol/L) ,释放到培养液中的谷氨酸的含量也相应增加 ;当培养液中去除钙离子时 ,细胞外液中谷氨酸的浓度不随氯化钾浓度的变化而改变。在含钙培养液中加入 4mg/ml的天麻素 ,再加入 10~ 5 0mmol/L氯化钾 ,未见细胞外液中的谷氨酸含量增高。结论 氯化钾诱导的SH SY5Y细胞谷氨酸的释放增多与细胞外液钙离子内流有关 ;天麻素可以颉颃由氯化钾诱导的神经细胞谷氨酸释放增加的作用。
Objective To investigate the effect of gastrodin on the release of glutamate from neurons induced by potassium chloride. Methods The content of glutamic acid in the culture fluid of human neuroblastoma SH SY5Y cell line was determined by high performance liquid chromatography (HPLC). RESULTS When calcium ions were present in the extracellular fluid, the content of glutamic acid released into the culture fluid increased with increasing potassium chloride concentration (10-30 mmol/L); when calcium ions were removed from the culture fluid The concentration of glutamate in the extracellular fluid does not change with the concentration of potassium chloride. In the culture medium containing calcium, 4 mg/ml of gastrodin was added, and then 10 to 50 mmol/L of potassium chloride was added. No increase in glutamate content in the extracellular fluid was observed. Conclusion The increase of glutamate release induced by potassium chloride in SH SY5Y cells is related to the extracellular calcium influx. Gastrodin can increase the release of glutamate from neurons induced by potassium chloride.
