目的探讨α-细辛醚对FMR1基因敲除小鼠的旷场行为的干预作用。方法通过对30日龄FMR1基因敲除小鼠连续腹腔注射不同剂量α-细辛醚5天,用药第5天进行旷场行为实验,观察能否改善KO鼠过度活动的表型。结果在旷场行为实验中,与WT鼠相比,未使用α-细辛醚的KO鼠的总运动长度及总跨格次数,中央格(区4)活动时间,中央格(区4)进入次数均比WT鼠多,P<0.05;使用α-细辛醚后,KO鼠α-细辛醚6~24 mg/kg剂量组与生理盐水(0 mg/kg)组比较,旷场实验运动长度及跨格次数均有减少(P<0.05)。结论 6~24mg/kgα-细辛醚能改善KO鼠活动过度的表型,可能对FMR1基因敲除小鼠有治疗作用。
Objective To investigate the effects of α-asarone on the open-field behavior of FMR1 knockout mice. Methods The 30-day-old FMR1 knockout mice were injected intraperitoneally with different doses of α-asarone for 5 days. The open field behavioral test was performed on the 5th day of treatment to observe whether the over-activity phenotype could be improved in KO mice. Results In the open-field behavioral test, compared with WT rats, the total exercise length and total number of cross-sections of KO rats without α-asarone, the activity time of the middle cell (area 4), the middle cell (area 4) (P <0.05). Compared with the normal saline (0 mg / kg) group, the dose of α-asarone in the KO rat 6-24 mg / kg group was significantly higher than that of the WT group Length and cross-grid number decreased (P <0.05). Conclusion 6 ~ 24mg / kg α-asarone can improve the hyperactivity phenotype in KO rats, which may have a therapeutic effect on FMR1 knockout mice.
