GGl_4对肝脏的毒性可能是由于它在肝脏中形成了活性代谢产物 GGl_3游离基。该游离基一方面可与不同的细胞成分反应,另一方面又可引起内质网的不饱和聚脂类的过氧化作用。作者用 Sprague-Dawley 雄性大鼠(250~280g)分别腹腔注射20%GGl_4橄榄油溶液(5ml/kg 体重)及~(14)GGl_4(14×16~6 dpm/ml)橄榄油溶液(5ml/kg 体重),对照组只注射橄榄油。给 GGl_4后分别在3、6小时处死动物。测定睾丸中的微粒体酶 P-450含量和~(14)GGl_4中的~(14)G与肝脏和睾丸的微粒体脂类的不可逆结合和GGl_4所引起的该脂类的过氧化作用。
GGl_4 toxicity to the liver may be due to its formation of the liver in the active metabolite GGl_3 free radicals. This radical, on the one hand, can react with different cellular components and, on the other hand, can cause peroxidation of the endoplasmic reticulum unsaturated polyesters. In this study, Sprague-Dawley male rats (250-280 g) were intraperitoneally injected with 20% GGI_4 olive oil (5 ml / kg body weight) and ~ (14) GGl_4 (14 × 16-6 dpm / ml) olive oil solution kg body weight), the control group only injected olive oil. Animals were sacrificed at 3 and 6 hours after GGl_4 administration. The content of microsomal enzyme P-450 in testes and the irreversible binding of ~ (14) G in ~ (14) GGl_4 to the microsomal lipids in liver and testis and the lipid peroxidation induced by GGl_4 were determined.
