目的研究胰石蛋白/再生蛋白(pancreatic stone protein/regenerating protein, PSP/reg)基因在急性胰腺炎(acute pancreatitis,AP)动物模型胰腺组织中的表达特点及意义。方法大鼠轻型胰腺炎(mild acute pancreatitis,MAP)和重型胰腺炎(severe acute pancreatitis,SAP)模型由逆行胰胆管注射1.5%及3.0%牛磺胆酸钠制备。采用逆转录聚合酶链反应(RT-PCR)法检测胰腺组织PSP/reg mRNA和TGF-β1 mRNA的表达,同时检测胰腺组织病理改变。结果AP诱导后24 h胰腺组织损伤最为严重,48 h后逐渐减轻。PSP/reg mRNA在正常胰腺组织呈低水平表达,AP诱导后24 h明显升高,于胰腺组织损伤减轻时(48 h)达到峰值,在AP恢复阶段呈持续高水平表达,与TGF-β1 mRNA的表达时相具有相似性,且SAP组明显高于MAP组(P<0.05)。结论PSP/reg可能与AP时胰腺组织损伤后的修复、再生有关。
Objective To study the expression and significance of pancreatic stone protein / regenerating protein (PSP / reg) gene in pancreatic tissue of acute pancreatitis (AP) animal model. Methods Rats with mild acute pancreatitis (MAP) and severe acute pancreatitis (SAP) were prepared by injecting 1.5% and 3.0% sodium taurocholate into the pancreatic duct. The expression of PSP / reg mRNA and TGF-β1 mRNA in pancreatic tissues were detected by reverse transcription-polymerase chain reaction (RT-PCR), and the pathological changes of pancreatic tissues were also detected. Results The pancreatic tissue injury was the most serious at 24 h after AP induction, and gradually decreased after 48 h. PSP / reg mRNA was expressed at low level in normal pancreas. The level of PSP / reg mRNA was significantly increased at 24 h after AP induction and peaked at 48 h after injury of pancreatic tissue. The expression of PSP / reg mRNA was consistently high at the stage of AP recovery, (P <0.05), and the SAP group was significantly higher than MAP group (P <0.05). Conclusion PSP / reg may be related to the repair and regeneration of pancreatic tissue after AP injury.
