多糖是白僵蚕的重要活性成分之一。采用水提醇沉法提取白僵蚕的多糖组分,在单因素试验的基础上,选择提取温度、提取时间和原料质量浓度3个因素设计Box-Benhnken中心组合试验,以白僵蚕多糖得率为响应值进行响应面分析优化提取工艺条件,并考察白僵蚕多糖提取物的体外抗氧化活性。优化的白僵蚕多糖提取工艺条件为提取温度100℃、提取时间160 min、原料质量浓度40 g/L,在此条件下白僵蚕多糖的得率为4.43%,提取液经醇沉干燥后所得粗多糖纯度为49.22%。获得的白僵蚕多糖提取物具有较好的抗氧化活性,且总还原力和清除羟自由基能力均呈现一定的量效关系。建立的白僵蚕多糖提取工艺操作简单、提取效果较好,白僵蚕多糖的抗氧化活性有利于白僵蚕在中成药或功能食品方面的深度开发利用。
Polysaccharide is one of the important active ingredients of white silkworm. The polysaccharide composition of white silkworm (Bombyx mori) was extracted by water extraction and alcohol precipitation. Based on the single factor test, three factors of extraction temperature, extraction time and raw material concentration were selected to design Box-Benhnken central combination test. Rate of response surface analysis to optimize the extraction process conditions, and to observe the white silkworm polysaccharide extract in vitro antioxidant activity. The optimum extraction conditions of white silkworm polysaccharides were as follows: the extraction temperature was 100 ℃, the extraction time was 160 min, the mass concentration of raw materials was 40 g / L, the yield of polysaccharide was 3.43% The crude polysaccharide obtained had a purity of 49.22%. The obtained polysaccharides of white silkworm polysaccharide had good antioxidant activity, and the total reducing power and ability of scavenging hydroxyl radical showed a certain dose-effect relationship. The established polysaccharides extracted from white silkworm silkworm has simple operation and good extraction effect. The antioxidant activity of white silkworm polysaccharide is conducive to the in-depth development and utilization of white silkworm in proprietary Chinese medicines or functional foods.
