目的:视网膜缺血是临床常见病之一,目前有关视网膜不完全性缺血情况下细胞凋亡和凋亡相关基因表达的尚未见研究报告.方法:通过结扎双侧颈总动脉造成大鼠视网膜不完全性缺血1、3、5、14、30、60天,用TUNEL方法标记视网膜凋亡细胞,并用体视学方法进行定量分析,用免疫组化方法观察bcl-2的表达情况.结果:1.缺血1天节细胞层和内核层即见凋亡细胞,以内核层为多;3天在外核层也出现.7天与3天无明显差别.14天凋亡细胞增多,达高峰,术后30及60天凋亡细胞的数密度与14天无明显差异.2.缺血1天Muller’s细胞内侧部bc-2表达增强,包括终足及位于节细胞层的突起.内界膜处着色明显,节细胞层出现“斑、点、线”状和一些细胞样阳性着色结构.3天和7天,着色区域扩大至Muller’s细胞外侧部,在外界膜呈现一条着色线,外核层出现垂直走向的着色纤维,外网层出现着色网.14天bcl-2阳性反应局限于Muller’s细胞内侧部,节细胞层内细胞样着色团块增多,Muller’s细胞外侧部着色不明显.缺血60天,bcl-2表达明显
