目的 探讨缓激肽 ( BK)对大鼠尾动脉的缩血管作用机制。方法 对离体正常大鼠尾动脉用 Biopac系统进行测定 ,观察 BK对血管张力的影响 ;分离平滑肌细胞用膜片钳记录钙通道电流 ,观察BK对单个平滑肌细胞的影响。结果 BK可引起大鼠尾动脉收缩且呈量效关系 ,细胞内 Ca2 +和细胞外Ca2 +在其中发挥了重要作用 ,且不受内皮细胞的影响 ;KCl引起细胞膜去极化后加强了 BK的收缩作用。结论 BK在大鼠尾动脉的缩血管作用是细胞内钙和细胞外钙依赖性的。
Objective To investigate the vasoconstriction mechanism of bradykinin (BK) on rat tail artery. Methods The tail artery of isolated normal rats were measured with Biopac system to observe the effect of BK on vascular tone. Smooth muscle cells were isolated and the calcium channel current was recorded by patch-clamp. The effect of BK on single smooth muscle cells was observed. Results BK induced contraction of rat tail artery in a quantitative and dose-dependent manner. Intracellular Ca2 + and extracellular Ca2 + played an important role in them, and were not affected by endothelial cells. KCl caused depolarization of cell membrane and enhanced BK Shrinkage. Conclusions The vasoconstrictor effect of BK in rat tail artery is intracellular calcium and extracellular calcium-dependent.
