目的揭示不同病程2型糖尿病(T2DM)肾病大鼠肾动脉上大电导钙激活钾通道基因表达是否有改变。方法采用单肾切除+小剂量链脲佐菌素(streptozotocin,STZ)+高脂饮食复制T2DM大鼠肾病模型,采用实时定量聚合酶链反应(PCR)法测定肾动脉BK_(Ca)通道的ɑ、β1亚单位的mRNA表达水平,同时全自动血生化仪检测血尿素氮、血肌酐;收集24 h尿蛋白定量;计算肾脏指数;肾组织切片光、电镜观察病理改变。结果 8、12周T2DM肾病大鼠血糖,血胰岛素,血肌酐,尿素氮,24 h尿蛋白,肾脏肥大指数均明显高于对照组(P<0.05、P<0.01),大鼠肾脏病理形态变化明显。8周、12周T2DM肾病大鼠BK_(Ca)ɑ亚单位在肾动脉mRNA表达变化不明显;β1亚单位在肾动脉中表达下降(P<0.05、P<0.01)。结论 T2DM肾病大鼠肾动脉上BK_(Ca)通道β1亚单位mRNA表达降低,并与肾脏病变程度一致。
Objective To reveal whether there is any change in the gene expression of the large-conductance calcium-activated potassium channel in renal artery of type 2 diabetic rats with different course of disease. Methods The nephropathy model of T2DM rats was established by single nephrectomy + low dose streptozotocin (STZ) + high fat diet. The renal artery BK Ca channels were measured by real time quantitative polymerase chain reaction (PCR) , Β1 subunit mRNA expression levels, while automatic blood biochemical analyzer detection of blood urea nitrogen, serum creatinine; 24h urine protein quantitation; calculate the renal index; kidney tissue sections light, electron microscopy pathological changes. Results The blood glucose, blood insulin, serum creatinine, urea nitrogen, 24 h urine protein and renal hypertrophy index in T2DM nephropathy rats at 8 and 12 weeks were significantly higher than those in control group (P <0.05, P <0.01) obvious. The expression of BK_ (Ca) ɑ subunit in renal tissue of rats with T2DM at 8 weeks and 12 weeks was insignificant, while the expression of β1 subunit was decreased in renal arteries (P <0.05, P <0.01). Conclusion The expression of β1 subunit mRNA of BK_ (Ca) channel in renal artery of T2DM nephropathy rats is decreased, which is consistent with the degree of renal disease.
