目的:观察大鼠深Ⅱ度烫伤创面愈合过程中基质金属蛋白酶-2(matrix metalloproteinase 2,MMP-2)、基质金属蛋白酶-7与金属蛋白酶组织抑制因子-2(tissue inhibitor of metalloproteinase-2,TIMP-2)的变化以及创面外用bFGF 对两者的影响,认识基质金属蛋白酶与抑制因子对创面愈合的作用。方法:78只 Wistar 大鼠随机分为正常对照、单纯烫伤和 bFGF 治疗三组。利用大鼠30%深Ⅱ度烫伤模型,于伤后3h、6h、1d、3d、7d 和14d 采取创面皮肤标本,用免疫组织化学染色检测创面组织真皮内成纤维细胞 MMP-2、MMP-7和 TIMP-2的表达变化情况。结果:烫伤大鼠创面中细胞外基质活动影响皮肤的胶原沉积。伤后1d MMP-2/MMP-7表达增多,随后略有下降,7d 时再次达到高峰。伤后 TIMP-2的表达与前者的表达一致。局部外用 bFGF 后,创伤初期(3h~6h)组织中 MMP-2/MMP-7的表达无显著变化,1d~14d,MMP-2/MMP-7和 TIMP-2阳性表达强于对照组。结论:MMPs 和 TIMPs 的表达变化是组织修复的重要步骤,bFGF 加速创面愈合的过程与 MMP-2/MMP-7和 TIMP2的表达密切相关。
Objective: To observe the expression of matrix metalloproteinase-2 (MMP-2), matrix metalloproteinase-7 (TIMP-2) and tissue inhibitor of metalloproteinase-2 -2) as well as the effect of topical bFGF on wound healing, and to understand the effect of matrix metalloproteinases and inhibitors on wound healing. Methods: Totally 78 Wistar rats were randomly divided into normal control group, scald group and bFGF treatment group. The wounds were collected at 3h, 6h, 1d, 3d, 7d and 14d after wound healing in rats with 30% deep second degree scald. The expressions of MMP-2 and MMP-7 in wound dermal fibroblasts were detected by immunohistochemical staining And TIMP-2 expression changes. Results: Extracellular matrix activity in scalded rat wounds affected collagen deposition in the skin. The expression of MMP-2 / MMP-7 increased on the 1st day after injury, then decreased slightly and peaked again on the 7th day. After injury, the expression of TIMP-2 was consistent with that of the former. After topical application of bFGF, the expression of MMP-2 / MMP-7 had no significant change in the early stage of trauma (3h ~ 6h). The process of bFGF accelerating wound healing is closely related to the expression of MMP-2 / MMP-7 and TIMP2.
