对7例正常前列腺和16例良性增生前列腺(BPH)以机械法分离上皮和间质细胞,以氚标睾酮为底物,以TLC法分离酶催化产物,依二氢睾酮(DHT)生成量测定5α-还原酶活性,结果显示:BPH和正常组织间质和上皮部分的酶活性[(pmol DHT/(mg蛋白质·30min)]分别为:91.4±18.1和28.6±7.4及44.7±8.9和23.9±6.8,表明5α-还原酶主要集中在间质部分,BPH组织间质部分酶活性增高,提示5α-还原酶的分布及活性改变与BPH的发生有密切关系。
Epithelial and interstitial cells were mechanically isolated from 7 normal prostate and 16 benign prostatic hyperplasia (BPH). Enzymatic products were separated by TLC using tritiated testosterone as a substrate, and dihydrotestosterone (DHT) production was measured 5α-reductase activity, the results showed that: (pmol DHT / (mg protein · 30min)] of 91.4 ± 18.1 and 28.6 ± 7.4 and 44.7 ± 8.9 and 23.9 ± 6.8, indicating that 5α-reductase mainly in the interstitial part of the BPH tissue interstitial part of the enzyme activity, suggesting that 5α-reductase distribution and activity changes are closely related with the occurrence of BPH.
