目的 探讨外源性血小板源伤口愈合因子 (platelet derivedwoundhealingfactors,PDWHF)改善糖尿病难愈性伤口愈合的作用与 β -转化生长因子 (TGF - β)基因表达的关系。 方法 44只雄性SD大鼠 ,分为正常对照组 (鼠数 =12 )、糖尿病组 (鼠数 =32 )。四氧嘧啶诱导糖尿病组大鼠血糖值 >1.8g/L 1~ 2d后 ,在每组大鼠背部用外科方法剪去 2块直径为 1.8cm的全层皮肤 ,造成全皮层开放伤口。术后当日及以后连续 6日 ,选糖尿病大鼠一侧伤口为PDWHF治疗组伤口 ,在其局部应用PDWHF ( 10 0 μg/伤口 ) ,另一侧 (糖尿病对照组伤口 )及正常对照组伤口不予PDWHF治疗。 结果 伤后 5d ,PDWHF治疗组伤口与糠尿病对照组伤口闭合指数无差异 ,均低于正常对照组 (P <0 .0 5 ) ;伤后 7,10 ,14d ,PDWHF治疗组伤口闭合程度明显高于糠尿病对照组伤口(P <0 .0 5 ) ,但仍低于正常对照组 (P <0 .0 5 )。伤后 5 ,7d ,PDWHF治疗组伤口组织中TGF - βmRNA水平量是糖尿病对照组的 4倍和 5 .6倍 (P <0 .0 1) ,但低于正常对照组 (P <0 .0 5 ) ;伤后 10d ,3组间TGF - βmRNA水平量无明显差异。 结论 PDWHF促糖尿病大鼠伤口愈合与其增强伤口组织TGF - β基因表达相关。
Objective To investigate the relationship between the effect of exogenous platelet derived wound healing factor (PDWHF) on the healing of diabetic refractory wound and the expression of β - transforming growth factor (TGF - β) gene. Methods Forty-four male SD rats were divided into normal control group (the number of mice = 12) and diabetic group (the number of rats = 32). Conclusion PDWHF induces wound healing in diabetic rats and enhances the expression of TGF - β gene in wound tissues.
