目的 观察高温顺铂肢体区域性隔离灌注的局部和全身不良反应。方法 12只青紫兰兔随机分成两组 ,在相同灌注条件下 ,一组用DDP(2mg/kg体重 )灌注 ,另一组用生理盐水灌注。灌注前及灌注后第 3天测定相关血生化 ,电解质及白细胞计数。观察肢体感染、水肿、运动等功能。根据Wiberdinks分级评价局部不良反应等级。另 1只动物按 4mg/kg体重DDP灌注 ,观察上述指标。 结果 在灌注及随访期间无动物死亡。两组实验动物灌注前后AST、AKP、Cr,电解质无明显差异。实验组LDH ,CK灌注后有显著升高。ALT、Urea、白细胞在DDP组与灌注前有差异 ,前者高于后者。除轻度胃肠道反应外 ,无严重肝肾功能受损 ,骨髓抑制等不良反应。局部肢体毒性分级 ,DDP组明显高于无药组。按 4mg/kg体重灌注的动物出现明显的全身及局部不良反应。 结论 用高温 2mg/kg顺铂肢体区域性灌注无全身严重并发症 ,局部不良反应也较轻 ,因而是一种较为安全的治疗方法
Objective To observe the local and systemic adverse reactions of high-temperature cisplatin limb regional isolated perfusion. Methods Twelve green rabbits were randomly divided into two groups. Under the same infusion conditions, one group was infused with DDP (2 mg/kg body weight), and the other group was infused with normal saline. Correlative blood biochemistry, electrolyte and white blood cell counts were measured before and 3 days after perfusion. Observe limb infection, edema, exercise and other functions. The level of local adverse reactions was evaluated according to the Wiberdinks grading. The other animal was infused with 4 mg/kg body weight DDP and the above indicators were observed. Results No animal died during perfusion and follow-up. There was no significant difference in electrolytes of AST, AKP, Cr between the two groups before and after perfusion. LDH in the experimental group was significantly increased after CK perfusion. ALT, Urea, and WBC differed between the DDP group and the perfusion group. The former was higher than the latter. In addition to mild gastrointestinal reactions, there was no serious liver and kidney dysfunction, bone marrow suppression and other adverse reactions. Local limb toxicity grade was significantly higher in the DDP group than in the no-drug group. Animals infused at 4 mg/kg body weight showed significant systemic and local adverse reactions. Conclusions There is no serious systemic complications in the regional perfusion of high-temperature 2mg/kg cisplatin, and the local adverse reactions are also lighter, so it is a safer treatment method.
