目的:观察平阳霉素诱发的大鼠白内障模型晶体上皮细胞DNA损伤及修复的动态过程。方法:单细胞电泳法(Singlecellgelassay,SCG)。结果:首次注射平阳霉素12h即出现严重的DNA单链断裂(DNAsinglestrandbreaks,SSB),直至白内障产生,SSB仍很严重,该DNA损伤较其它模型更严重,其晶体囊内容物在致障过程中出现明显的液化与崩解。结论:平阳霉素性白内障模型在致障过程中有较其它模型更严重的DNA单链损伤,且可能有不尽相同的机理存在。
Objective: To observe the dynamic process of DNA damage and repair of lens epithelial cells induced by Pingyangmycin in rat cataract model. Methods: single cell electrophoresis (Singlecellgelassay, SCG). Results: After the first injection of pingyangmycin, severe DNA single strand breaks (SSBs) appeared, and until the cataract was produced, the SSB was still serious. CONCLUSION: Pingyangmycin cataract model has more severe single-strand DNA damage than other models in the process of catastrophe, and may have different mechanisms.
