分别对来自比利时和荷兰的水曲柳原木进行了病原菌分离,结果在原木的树皮中各分离到1株疑似苹果壳色单隔孢溃疡病菌的菌株Bs-1268和Bs-8212。对其进行了病原菌形态学观察、致病性测定并结合分子生物学方法检测,实验结果表明,菌株Bs-1268和Bs-8212在PDA和OA培养基上均能产生一定量的分生孢子器和分生孢子,分生孢子器通常为球形;经真菌通用引物(ITS4/ITS5)扩增和测序,菌株Bs-1268和Bs-8212与Gen Bank中登录号KF766158.1的菌株序列同源性达到100%;病菌分别接种苹果和梨6 d后发病。根据上述实验结果,将分离获得的菌株Bs-1268和Bs-8212鉴定为苹果壳色单隔孢溃疡病菌(Botryosphaeria stevensii Shoemaker)。
Pathogenic bacteria were isolated from the logs of the wild ash from Belgium and the Netherlands respectively. As a result, one isolate Bs-1268 and Bs-8212 were isolated from the bark of log respectively. Morphological observation, pathogenicity determination and molecular biological methods were used to detect the pathogen. The results showed that strain Bs-1268 and Bs-8212 could produce a certain amount of conidia on PDA and OA medium And conidia, the conidia were usually spherical. The sequences of the strains Bs-1268 and Bs-8212 were identical with those of GenBank accession number KF766158.1 by universal fungal primers (ITS4 / ITS5) Reaching 100%; pathogen were inoculated with apple and pear disease after 6 d. Based on the above experimental results, the strains Bs-1268 and Bs-8212 obtained from isolation were identified as Botryosphaeria stevensii Shoemaker.
