目的探讨编码人类中性粒细胞抗原-3(Human neutrophil antigens-3,HNA-3)的SLC44A2基因在江苏省汉族人群、美国白种人及美国黑人中的多态性及频率分布。方法采用序列特异性引物聚合酶链反应(Polymerase chain reaction-sequence specific priming,PCR-SSP)方法,分别对随机无亲缘关系的3个种族献血者的SLC44A2基因进行检测,其中包括江苏省的203名汉族、美国的102名白种人和245名黑人;同时采用PCR方法扩增相关外显子及侧翼内含子并进行测序分析,进一步验证PCR-SSP结果的准确性。结果汉族人群中HNA-3a/3a有91例,HNA-3a/3b 86例,HNA-3b/3b 26例;美国白种人中HNA-3a/3a有73例,HNA-3a/3b有26例,HNA-3b/3b有3例;美国黑人中HNA-3a/3a有212例,HNA-3a/3b有31例,HNA-3b/3b有2例。HNA-3a在江苏汉族人群,美国白种人及黑人中的基因频率分别为0.66,0.84和0.93;HNA-3b在江苏汉族人群,美国白种人及黑人中的基因频率分别为0.34,0.16和0.07。HNA-3b/3b纯合子在江苏省汉族人群的频率明显高于美国白种人及美国黑人,具有统计学意义(P<0.01)。结论编码HNA-3抗原的等位基因频率在3种不同种族中的分布存在多态性;该抗原的多态性及可能造成的TRALI需要引起临床上更多的关注。
Objective To investigate the polymorphism and frequency distribution of SLC44A2 gene encoding human neutrophil antigens-3 (HNA-3) in Jiangsu Han population, American Caucasian and African American. Methods The SLC44A2 gene of 3 unrelated unrelated donors was detected by polymerase chain reaction-sequence specific priming (PCR-SSP), including 203 in Jiangsu Province Han Chinese, the United States of America 102 white and 245 black people; at the same PCR amplification of related exons and flanking introns and sequencing analysis to further verify the accuracy of PCR-SSP results. Results There were 91 cases of HNA-3a / 3a, 86 cases of HNA-3a / 3b and 26 cases of HNA-3b / 3b in Han population. There were 73 cases of HNA-3a / 3a and 26 cases of HNA-3a / 3b For example, there are 3 cases of HNA-3b / 3b, 212 cases of HNA-3a / 3a in African American, 31 cases of HNA-3a / 3b and 2 cases of HNA-3b / 3b. The gene frequencies of HNA-3a in Jiangsu Han population and American Caucasian and black population were 0.66,0.84 and 0.93, respectively. The frequencies of HNA-3b gene in Jiangsu Han population, American Caucasian population and black population were 0.34,0.16 and 0.07. The frequency of HNA-3b / 3b homozygotes in Han nationality in Jiangsu Province was significantly higher than that in white Americans and African Americans (P <0.01). Conclusion The frequency of allele encoding HNA-3 antigen is polymorphic in three different races. The polymorphism of the antigen and possible TRALI needs more clinical attention.
