应用薄层荧光扫描法测定了动物血浆中佐匹克隆的浓度,并对佐匹克隆在中毒家兔体内的分布进行了研究。样品在pH686条件下用二氯甲烷—异戊醇(98∶2)提取,氯仿—甲醇(9∶1)展开,用CS—930薄层扫描仪在激发波长305nm进行荧光单波长线性扫描测定。本法对血浆中佐匹克隆的最低检出限为25ng/mL,平均提取回收率80%,RSD为204%。研究发现,急性中毒家兔12h后其胃内容物、肝、脑和胆汁中佐匹克隆含量较高。
The concentration of zopiclone in animal plasma was determined by TLC fluorescence scanning and the distribution of zopiclone in rabbits was studied. The sample was extracted with dichloromethane-isoamyl alcohol (98: 2) at pH6.86 and developed with chloroform-methanol (9: 1). Fluorescence single wavelength linear scanning was performed at a excitation wavelength of 305 nm with a CS-930 TLC scanner Determination. The detection limit of this method for zopiclone in plasma is 25 ng / mL, with an average recovery of 80% and a RSD of 2.04%.
