高脂饲料喂养大鼠 ,诱发胰岛素抵抗形成血糖升高 ,以研究瘦素与糖尿病的关系。将大鼠分为正常和高脂饲料喂养组 ,于喂养前及喂养 30、6 0d时测血糖、瘦素与胰岛素水平 ,行高胰岛素正常血糖钳夹试验 ,最后分离脂肪称重。结果 :高脂饮食组具有明显的胰岛素抵抗 ,有 5 0 %血糖升至 8 3mmol L以上 ;形成糖尿病的大鼠体重与对照组无明显区别 ;高脂饮食组瘦素与胰岛素水平与对照组比较有明显差异 ;所有大鼠瘦素水平与体重、胰岛素、血糖均呈正相关。结论 :(1)高脂饮食是导致胰岛素抵抗形成糖尿病的诱因之一 ;(2 )体脂含量与糖尿病正相关 ;(3)瘦素抵抗与胰岛素抵抗形成糖尿病的发病机制可能有关 ;(4)瘦素水平随着鼠龄的增加而增高 ,且参与体重的调节
Rats fed high-fat diet inducing insulin resistance to form hyperglycemia to study the relationship between leptin and diabetes. The rats were divided into normal and high-fat diet group, blood glucose, leptin and insulin levels were measured before feeding and 30, 60 days after feeding, and hyperinsulinemic normal blood glucose clamp test was performed. Finally, the fat was separated and weighed. Results: The high-fat diet group had obvious insulin resistance, 50% blood sugar rose to more than 8 3mmol L; the body weight of diabetic rats was no significant difference with the control group; high-fat diet group leptin and insulin levels compared with the control group There was a significant difference; all rats leptin level and body weight, insulin, blood glucose were positively correlated. (2) body fat content is positively correlated with diabetes; (3) leptin resistance may be related to the pathogenesis of insulin resistance and diabetes mellitus; (4) high fat diet may be one of the reasons leading to the formation of diabetes mellitus with insulin resistance; Leptin levels increase with increasing age, and participate in the regulation of body weight
