目的:探讨原发性高血压(essential hypertension,EH)患者单核细胞中经典瞬时受体电位通道3(transient receptorpotential canonical channel 3,TRPC3)转录水平与单核细胞中白细胞介素1β(interleukin-1β,IL-1β)和肿瘤坏死因子-α(tumornecrosis factor-α,TNF-α)的转录水平以及外周血单核细胞趋化蛋白1(monocyte chemotactic protein 1,MCP-1)浓度的关系。方法:抽取20例原发性高血压患者和20例血压正常的健康体检者的静脉血,分离获得单核细胞,运用实时定量聚合酶链反应法(re-al-time PCR)分别检测外周血单核细胞中TRPC3、TNF-α和IL-1β的mRNA,用酶联免疫吸附法(enzyme linked immunosorbentassay,ELISA)检测外周血MCP-1浓度。结果:与血压正常对照组比较,原发性高血压组单核细胞中TRPC3、TNF-α和IL-1β的mRNA表达升高,MCP-1浓度增高,具有统计学差异(P<0.05)。单核细胞中TRPC3的mRNA水平与TNF-α和IL-1β的mRNA水平以及MCP-1浓度呈现正相关。结论:原发性高血压作为一个慢性炎症过程,单核细胞中TRPC3表达与细胞因子水平相关,TRPC3表达上调可被认为是单核细胞活化的指标之一。
OBJECTIVE: To investigate the transcriptional level of TRPC3 in monocytes of patients with essential hypertension (EH) and the level of interleukin-1β , IL-1β) and tumor necrosis factor-α (TNF-α) and the level of monocyte chemotactic protein 1 (MCP-1) Methods: Mononuclear cells were isolated from 20 patients with essential hypertension and 20 healthy subjects with normotensive blood samples. Peripheral blood was detected by real-time quantitative polymerase chain reaction (re-al-time PCR) The mRNA levels of TRPC3, TNF-α and IL-1β in mononuclear cells were detected by enzyme linked immunosorbentassay (ELISA). Results: Compared with normotensive subjects, the mRNA expression of TRPC3, TNF-α and IL-1β in monocytes of patients with essential hypertension increased and the concentration of MCP-1 increased, with statistical significance (P <0.05). TRPC3 mRNA levels in monocytes positively correlated with the mRNA levels of TNF-α and IL-1β and MCP-1 concentrations. Conclusion: Primary hypertension is a chronic inflammatory process. TRPC3 expression in monocytes is correlated with cytokine levels. Upregulation of TRPC3 may be considered as one of the indicators of monocyte activation.
