醒脑静注射液对脑缺再灌注损伤小鼠血清IL-6和TNF-α水平影响

目的探讨醒脑静注射液对脑缺再灌注损伤小鼠血清IL-6和TNF-α水平影响。方法选择60只健康昆明雄性小鼠,分为假手术组、模型组、高剂量组、中剂量组、低剂量组。假手术组仅分离颈总动脉,其他四组分离颈总动脉行缺血再灌注。模型制造成功后第2天,假手术组和模型组给予同体积生理盐水,而高中低剂量组分别给予醒脑静注射液,剂量分别为10ml/kg、5ml/kg、2.5ml/kg,稀释为0.5ml后腹腔注射。1次/d,连续注射液5d。各组小鼠在末次给药后12h后采用摘除眼球法取血,离心后取血清,采用放射免疫法测定小鼠血清IL-6和TNF-α水平。结果模型组IL-6和TNF-α水平分别高于假手术组,差异有统计学意义(P<0.05);高、中、低剂量组IL-6和TNF-α水平分别低于模型组,差异有统计学意义(P<0.05);高剂量组IL-6和TNF-α水平分别低于中剂量组,差异有统计学意义(P<0.05);中剂量组IL-6和TNF-α水平分别低于低剂量组,差异有统计学意义(P<0.05)。结论 IL-6和TNF-а在脑缺血再灌注损伤中表达水平升高,参与了脑缺血再灌注损伤,而醒脑剂注射液能够降低IL-6和TNF-α表达,对脑缺血再灌注损伤中的炎症反应有一定的抑制作用。 Objective To investigate the effects of xingnaojing injection on serum IL-6 and TNF-α in mice with cerebral ischemia-reperfusion injury. Methods Sixty male Kunming mice were selected and divided into sham operation group, model group, high dose group, middle dose group and low dose group. The sham operation group only separated the common carotid artery, the other four groups were separated from the common carotid artery for ischemia-reperfusion. On the second day after the successful manufacture of the model, sham-operation group and model group were given the same volume of normal saline, while the high-middle-low dose group were given Xingnaojing injection, the dose were 10ml / kg, 5ml / 0.5ml after intraperitoneal injection. 1 times / d, continuous injection 5d. The mice in each group were sacrificed at 12h after the last administration, and the serum was taken after centrifugation. The levels of IL-6 and TNF-α in serum were measured by radioimmunoassay. Results The levels of IL-6 and TNF-α in the model group were significantly higher than those in the sham operation group (P <0.05). The levels of IL-6 and TNF-α in the model group were lower than those in the model group (P <0.05). The levels of IL-6 and TNF-α in the high dose group were lower than those in the middle dose group, the difference was statistically significant (P <0.05) The levels were lower than the low dose group, the difference was statistically significant (P <0.05). Conclusion The expression of IL-6 and TNF-а in cerebral ischemia-reperfusion injury is increased, which is involved in cerebral ischemia-reperfusion injury. Xingnao injection can reduce the expression of IL-6 and TNF-α, Inflammatory reaction in blood reperfusion injury has a certain inhibitory effect.
