目的 :分析猪囊尾蚴 DNA疫苗 pc DNA3- c C1肌肉接种后在猪体内的组织分布。 方法 :分别于疫苗接种后 1d、7d、4周、8周 ,提取猪骨髓、肾、肝、脑、性腺、肠系膜淋巴结、脾、胸腺、外周血及肌注部位的肌肉和皮肤的总 DNA,通过 PCR的方法检测疫苗质粒在各组织中的分布及随时间变化的情况。 结果 :疫苗接种后的第 1天几乎所有组织中都检测到质粒 ;但随着时间的延续 ,仅在注射部位的肌肉和皮肤中检测到质粒的存在 ,并保持到 8周以上。 结论 :肌注疫苗质粒后 ,实验猪组织中的游离质粒在短时期内就被清除
OBJECTIVE: To analyze the tissue distribution of pig cysticercus DNA vaccine pc DNA3-c C1 in pigs after inoculation. Methods: The total DNA of muscle and skin of bone marrow, kidney, liver, brain, gonad, mesenteric lymph node, spleen, thymus, peripheral blood and intramuscular injection site were extracted at 1d, 7d, 4w and 8w after vaccination. The distribution of the vaccine plasmid in each tissue and the change with time were detected by PCR method. Results: Plasmids were detected in almost all tissues on day 1 after vaccination; however, over time, the presence of plasmids was detected only in muscle and skin at the injection site and remained above 8 weeks. Conclusion: After intramuscular vaccine, the free plasmids in experimental porcine tissues were cleared in a short time
