以甜高粱品种罗马和W452与普通高粱品种忻粱52不同生育时期的叶片和茎秆为材料,对总糖、蔗糖含量以及蔗糖合成酶(sucrose synthase,SS)基因表达规律进行研究。结果表明:在叶片和茎秆的整个生育期,蔗糖为总糖的主要组成成分,其含量与总糖含量的变化趋势一致;在叶片中,总糖和蔗糖含量从拔节期开始逐渐升高,在灌浆期达到最高,随后又下降,同时通过对SS基因表达规律研究发现,SS基因表达量先于蔗糖含量在开花期达到最高;在茎秆中,蔗糖含量占总糖含量的60%,甜高粱的糖分含量明显高于普通高粱,完熟期时糖分含量达到最高,而SS基因表达规律与在叶片中表达相似,开花期达到最高,随后有所下降。因此,通过研究糖分含量变化趋势与SS基因表达规律为甜高粱分子育种创造了条件。
The contents of total sugar and sucrose and the expression of sucrose synthase (SS) gene in leaves of sweet sorghum variety W452 and common sorghum variety Xin Liang 52 at different growth stages were studied. The results showed that sucrose was the main component of total sugar during the whole growth period of leaves and stems, and its content was consistent with the change trend of total sugar content. In the leaves, total sugar and sucrose content gradually increased from the jointing stage, At the same time, the SS gene expression was found to be the highest at flowering stage prior to sucrose content. In the stem, sucrose content accounted for 60% of the total sugar content, sweet Sorghum sugar content was significantly higher than ordinary sorghum, reached the highest sugar content at maturity, while the expression of SS gene expression similar to the leaves, flowering reached the highest, then declined. Therefore, by studying the changes of sugar content and SS gene expression rules for molecular breeding of sweet sorghum to create the conditions.
